There a number of reasons why IIS could fail to start, but could you kindly see if any of the following resolves the issue:
-Run the profiler *explicitly* as administrator (you may need to press shift as your right-click on the profiler to get the "run as administrator" option)
-Disable any antiviruses or firewalls
-Try profiling using "original port" (please note that this will restart IIS)
